T7 RNA Polymerase with Amber Codon
This composite part contains the coding sequence of the chromosomal copy of T7RNA Polymerase from the E. coli KRX strain (uniprot.org)combined with an araBAD promoter. This T7 RNA polymerase has an amber codon at the first postion after the methionin. The bacteriophage T7 RNA Polymerase is a DNA-dependend RNA Polymerase used to express a gene of interest which is under the control of the corresponding T7 promoter. Thus it does not interfere with E. coli's endogenous transcriptional system.
Usage and Biology
The coding sequence of the T7 RNA polymerase is under control of the araBAD promotor (BBa_K808000). This T7 RNA polymerase has an amber codon at position 1 of the coding sequence after the start codon. This part can be used for designing a possible positive selection plasmid. If the desired non-canonical amino acid is incorporated by amber codon supression the T7 RNA polymerase can be expressed and can in turn bind to a T7 promoter, e.g. in BBa_K2201404 for a postive selection based on fluorescence.
